DNA adductomics is the systematic analysis of large numbers of DNA adducts or
DNA modifications present in a particular DNA sample obtained at a specific point in
time. The resulting DNA adductomes are represented visually by “DNA adduct
fingerprint” adductome maps and adductome profiles that are unique to each DNA
sample. In combination, the map and profile interfaces serve to provide detailed
visualizations of DNA modification occurrence patterns and allow for comparisons
between multiple samples. Additionally, the identities of specific DNA modifications
revealed by the maps may be determined, e.g. DNA damage in the form of a DNA
adduct. The DNA adductome approach utilizes liquid chromatography coupled with
electrospray ionization tandem mass spectrometry (LC/ESI-MS/MS) and the
methodology is designed to detect the neutral loss of 2’-deoxyribose from positively
ionized 2’-deoxynucleoside adducts in multiple reaction monitoring mode (MRM)
transmitting the [M+H]+ > [M+H-116]+ transition over multiple transitions. Data analysis
is optimized and coupled with a comprehensive manual screening process designed to
minimize the number of artifactual DNA modifications that appear in the final analyses.
Identification of DNA adducts is carried out by comparison to authentic standards and by
stable isotope dilution methods.
