Abstract:
This chapter presents four ultrasensitive methods (as Hoechst and PicoGreen
versions) for the accurate quantification of non-repairable and repairable dsDNA
damage, as well as for the concentration of dsDNA. The assays can be applied to
genomic, mitochondrial and plasmid dsDNA. The first assay provides an accurate
quantification of dsDNA irrespective of size, and the following assays quantify (a) nonrepairable
and (b) repairable dsDNA damage: for case (a), two assays, one for the direct
quantification of small-sized (01000 bp) fragmented necrotic/apoptotic DNA, and the
other for the quantification of the 0-23 Kb smearing seen in a typical DNA agarose gel
electrophoresis; for case (b), a fourth assay that quantifies the percentage of DNA nicks
present in DNA samples.
